My understanding is that once you rent the room it becomes your place of residence and the Castle Doctrine kicks in and yes, you may protect yourself while there. Firearms can also be transported from the vehicle to the place of residence (room, in this case). What you can't do (legally) is go down for breakfast or hang out in any public area of the hotel, armed.
I've seen this in official print someplace.

For anyone else who wishes to post on this thread, please read through the other posts first. We’ve already established that you by law are allowed to carry to and fro from your room. And can have a firearm in your room as well.
