I do not believe they work on weekends or holidays but I am no expert. I do know, however, that updates they make in the system do not appear to you until the day after they are made. Therefore a status update they make on Friday will update for you to see on Saturday morning. Some updates such as "mailed" appear to automated and may show up even on Sundays (that others have reported).
